The structural, biochemical, and functional analysis of a protein requires that the protein be expressed and purified in a folded, functional form. A common method is to express the target protein fused to a tag, such as MBP (maltose binding protein). MBP is used for the expression and purification of recombinant proteins in three ways: improving translational expression, providing an affinity purification strategy, and enhancing the solubility of target proteins. An MBP-tagged protein is eluted from an amylose column with maltose, in a single step procedure by affinity chromatography.
